摘要: Schisandrae Fructus (SF), the dried fruit of Schisandra chinensis (Turcz.) Baill., is a well-known traditional herb used in Asia for enhancing physical work capacity as well providing anti-stress and anti-inflammatory effects. Extracts SF (SFe) have also been reported to increase skeletal muscle mass inhibit atrophy. We examined whether SFe had muscle-protective effects old mice after chronic forced exercises, and, if so, relevant mechanisms.Ten-month-old aged male were divided into six groups. One group received no swimming oral administration distilled water (Intact); other groups (SW), oxymetholone (OXY), or at 500, 250 125mg/kg (SFe500, SFe250, SFe125, respectively). Forced was conducted 2min 30min administration; treatment repeated 28 days. Muscle thickness, weight, lean proportion, strength examined. The sampled muscles subjected histopathological biochemical analyses. Plasma by analyses. The thicknesses calf gastrocnemius soleus, protein proportion increased significantly SW versus Intact, they further OXY SW. upregulated mRNA expression related synthesis (Akt1, PI3K) growth (A1R, TRPV4), while it downregulated mRNAs degradation (atrogin-1, MuRF1) inhibitor (myostatin, SIRT1). detected upregulation downregulation enhanced In addition, inhibited lipid peroxidation reactive oxygen species, accelerated activities endogenous anti-oxidants anti-oxidant enzymes. biochemistry showed decreases creatine, creatine kinase LDH SW, suggesting SFe. analyses revealed an myofibre diameter, immunohistochemistry increases myofibres immunoreactive ATPase apoptosis markers (caspase-3, PARP) oxidative stress (NT, 4HNE, iNOS).Oral SFe, especially SFe500, exercise-induced adaptive strengthening through anti-apoptotic effects, mediated via modulation gene degradation. These results suggest that may be helpful improvement various disorders adjuvant therapy exercise-based remedies.
