摘要: A series of human androgen receptor (AR) deletion mutants was constructed to study the relationship between various structural domains and their different functions in AR protein. Immunoblots wild type expressed COS-1 cells, revealed a doublet appearance all proteins. One exception an mutant lacking amino acid residues 51-211 that migrated as single protein band, possibly due altered post-translational modification. The steroid binding domain found be encoded by approx. 250 C-terminal end. Deletions truncations this part abolished hormone binding. N-terminal observed essential for transcriptional activation. large parts were transcriptionally inactive. Deletion yielded constitutively active protein, indicating absence displays inhibitory function. In ligand cells distributed over nucleus cytoplasm. addition directed receptors nucleus. contrast, DNA hinge region, almost exclusively cytoplasmic hormone. This lacks conserved homologous SV40 T- nucleoplasmin nuclear localization signal. Hormone induced transfer nucleus, presence second, dependent targeting mechanism.
