摘要: BACKGROUND: Tumor-derived cytokines play a significant role in the progression of head and neck squamous cell carcinoma (HNSCC). Targeting proteins, such as tristetraprolin (TTP), that regulate multiple inflammatory may inhibit HNSCC. However, TTP's cancer is poorly understood. The goal current study was to determine whether TTP regulates patients with HNSCC. METHODS: TTP messenger RNA (mRNA) protein expression were determined by quantitative real-time–polymerase chain reaction (Q-RT-PCR) Western blot analysis, respectively. mRNA stability cytokine secretion evaluated RT-PCR enzyme-linked immunoadsorbent assay, respectively, after overexpression or knockdown HNSCC tissue microarrays immunostained for interleukin-6 (IL-6) TTP. RESULTS: TTP lines found be inversely correlated IL-6, vascular endothelial growth factor (VEGF), prostaglandin E2 (PGE2). Knockdown increased cytokines. Conversely, cells led decreased VEGF, PGE2. Immunohistochemical staining IL-6 demonstrated intensity prognostic poor disease-specific survival (P = .023), tumor recurrence development second primary tumors .014), overall .019). CONCLUSIONS: The results down-regulation enhances promotes Furthermore, high biomarker prognosis. In much enhanced associated prognosis, therapeutic target reduce concurrently Cancer 2011. © 2011 American Society.
