Laser‐Activatible PLGA Microparticles for Image‐Guided Cancer Therapy In Vivo

摘要: Poly(lactide-co-glycolic acid) (PLGA) particles are biocompatible and bio­degradable, can be used as a carrier for various chemotherapeutic drugs, imaging agents targeting moieties. Micrometer-sized PLGA were synthesized with gold nanoparticles DiI dye within the shell, perfluorohexane liquid (PFH) in core. Upon laser irradiation, shell absorbs energy, activating core (liquid conversion to gas). The rapidly expanding gas is expelled from particle, resulting microbubble; this violent process cause damage cells tissue. Studies using cell cultures show that phagocytosed by single consistently vaporized energies of 90 mJ cm−2, destruction. Rabbits metastasized squamous carcinoma lymph nodes then evaluate anti-cancer effects these nodes. After percutaneous injection upon through optical droplet vaporization, ultrasound shows significant increase contrast comparison control. Histology electron microscopy confirm disrupted throughout nodes, which slows tumor growth rate. This study containing PFC liquids theranostic vivo.
