Xenon Enhances LPS-Induced IL-1β Expression in Microglia via the Extracellular Signal-Regulated Kinase 1/2 Pathway

摘要: The extracellular signal-regulated kinase (ERK) is involved in the cytokine production of immune cells. In this study, we show influence xenon on phosphatase activity, ERK 1/2 signalling and expression microglia. murine microglia cell line BV-2 was treated with 50 ng/ml lipopolysaccharide (LPS) 74% 21% O2 5% CO2. Cytokine levels were examined by gene analysis, Western blot enzyme-linked immunosorbent assay. Phosphatase inhibition assessed p-nitrophenylphosphate phosphorylation via blot. Xenon significantly enhanced LPS-mediated IL-1β expression. observed after or LPS treatment which inhibited use MEK inhibitor U0126. combination superimposed individual effects activation. decreased cellular activity 20% dephosphorylation up to 1 h. blocking reduced LPS-treated cells a level obtained alone. conclusion, LPS-induced activation signalling. Xenon’s interference phosphatases may be key feature affect multiple intracellular pathways.
