摘要: Abstract Glial cell line-derived neurotrophic factor (GDNF), a distant member of the transforming growth factor-β (TGFβ) family, is protein that essential for survival dopaminergic, motor, and peripheral neurons. To facilitate its study, we others have developed sensitive (low pg/ml) enzyme-linked immunosorbant assays (ELISA) to quantitate endogenous concentrations GDNF, along with neurotrophin-3 (NT-3) nerve (NGF). However, tissue levels GDNF in adult animals are not readily detected by ELISA do correlate well message RNA. Based upon previously described methods extraction TGFβ from samples, an acid-treatment procedure allow quantification total GDNF. This also was evaluated use when measuring NT-3 NGF biological samples. The increases detectable amounts NT-3, all samples most serum tested. Moreover, these values were as much 35 times greater than those using traditional buffers. Such elevated likely resulted acid treatment promoting dissociation ligands receptors or binding proteins, thereby making more analyte available be measured ELISA. These findings indicate appropriate sample measurement factors.
