摘要: Functional genomics has become a major focus in the study of microbial pathogenesis. This used functional genomic tool, differential display reverse transcription-PCR, to identify transcriptional profile Cryptococcus neoformans cells as they produced meningitis an immunosuppressed host. serial global gene expression during infection allowed for identification up- and down-regulated genes infection. During this profiling, single enzyme isocitrate lyase (ICL1) was found be up regulated at 1 week rabbit model time maximum host cellular response. The finding suggested that glyoxylate shunt pathway are important yeast's energy production However, site-directed icl1 mutants had no apparent virulence defect two animal models growth within macrophages. These observations suggest although yeast responded certain environmental cue(s) by increase ICL1 infection, not necessary progression C. Compounds specifically target only unlikely cripple vivo.
