摘要: In this study, eight commercially available, chemically defined Chinese hamster ovary (CHO) cell culture media from different vendors were evaluated in batch using an IgG-producing CHO DG44 line as a model. Medium adaptation revealed that the occurrence of even small aggregates might be good indicator growth performance subsequent high density cultures. Batch experiments confirmed medium has significant impact on bioprocess performance, but amino acid concentrations alone not sufficient to ensure superior and antibody production. However, some key acids limiting most could identified. Unbalanced glucose led cell-specific lactate ammonium production rates. media, persistently probably induced suppression respiration oxidative phosphorylation, known Crabtree effect, which resulted glycolysis rates along with continuous additional experiments, two basal supplemented feeds manufacturers six combinations, order understand combined metabolism fed-batch process. Cell growth, nutrient consumption metabolite rates, production, IgG quality detail. Concentrated feed supplements boosted almost threefold titers up sevenfold. Depending strategy, fourfold higher peak eightfold increased (up 5.8 g/L) achieved. The glycolytic flux was remarkably similar among fed-batches; however, substantially specific observed combinations. Further analysis addition additives, can make considerable contribution cells. glycosylation recombinant influenced by selection feeds. Differences 50 % monogalacto-fucosylated (G1F) mannose fraction observed.
