Bone marrow origin of a B-cell lymphoma

摘要: To search for precursors of the neoplastic B cells in a patient with nodular lymphoma, we produced monoclonal antibody to variable region idiotope on lymphoma IgM heavy chain. Clonal ancestors were identified by this marker among bone marrow pre- (5% 26%). A second antiidiotype (anti-Id) specific complete kappa recognized 10% and blood greater than 95% lymphomatous lymph nodes, including one obtained after tumor conversion diffuse large cell lymphoma. Immunoglobulin gene analysis surprisingly revealed expansion multiple clones early lineage marrow, members clone. The data suggest that arose through progression transformational events beginning marrow: first, creation an oligoclonal pool pre-B cells, subsequent single subclone into low grade homed node follicles, later more invasive form B-cell
