An improved cucumber mosaic virus-based vector for efficient decoying of plant microRNAs

摘要: We previously devised a cucumber mosaic virus (CMV)-based vector system carrying microRNA target mimic sequences for analysis of function in Arabidopsis thaliana. describe an improved version which cloning is achieved by annealing two partly-overlapping complementary DNA oligonucleotides insertion into infectious clone CMV RNA3 (LS strain) fused to the cauliflower virus-derived 35S promoter. LS-CMV variants were generated co-infiltrating plants with Agrobacterium tumefaciens cells harboring engineered RNA1 and RNA2 clones. The utility using agroinfection deliver LS-CMV-derived was demonstrated miR165/166 three solanaceous hosts: Nicotiana benthamiana, tobacco (N. tabacum), tomato (Solanum lycopersicum). In all hosts induced marked changes developmental phenotype. Inhibition miRNA accumulation increased mRNA (HD-ZIP III) tomato. Thus, CMV-derived delivered via simple, cheap powerful means launching virus-based mimics likely be useful high-throughput investigation wide range plants.
