摘要: MicroRNAs (miRNAs) play key roles in plant development and defense against pathogens. To establish the function of individual miRNAs, gain-of-function analysis is usually done by overexpressing a specific miRNA transgenic plants has proved very effective. Loss-of-function target mimic method now also increasingly being used. The mimics expressed sequester lead to changed phenotypes that elucidate function. However, it takes time obtain plants. avoid using plants, we have developed virus-based suppression system (VbMS) based on Tobacco rattle virus vector. sequences miR156, miR319, or miR164 were introduced into viral genomic RNA, which was then inoculated Arabidopsis thaliana resulting consistent with previous reports from expression targets miRNAs increased showing activity had been inhibited. VbMS here validated for loss-of-function Moreover, since only simple agroinfiltration rather than transformation needed, suitable large-scale approaches
