Improved particle-packed HPLC/MS microchips for proteomic analysis

摘要: The influence of packing process parameters (packing pressure, application ultrasound) and the stationary phase particle size (3.5 5 μm) on chromatographic performance HPLC/MS chips was systematically investigated for proteomic samples. First, reproducibility detection limits separation were evaluated with a low-complexity sample tryptic BSA peptides. adsorbent quality protein identification then tested typical proteomics high complexity, human plasma fraction (Cohn IV-4). All provided highly reproducible separations these samples, but improved conditions smaller sizes resulted in chromatograms narrower peaks correspondingly higher signal intensities. Improved increased peak capacity, number identified peptides, thus sequence coverage particularly low amounts.
