Proteomic View of Basement Membranes from Human Retinal Blood Vessels, Inner Limiting Membranes, and Lens Capsules

摘要: Basement membranes (BMs) are extracellular matrix sheets comprising the laminins, type-IV collagens, nidogens, and heparan sulfate proteoglycans, perlecan, collagen XVIII, agrin. In intact BMs, BM proteins physiologically insoluble partially resistant to proteolytic digestion, making BMs a challenge study. Here three types of from adult human eyes, inner limiting membrane (ILM), retinal vascular lens capsule, were isolated for analysis by 1D-SDS-PAGE LC-MS/MS. Peptide protein identifications done using MaxQuant. 1129 identified with 1% false discovery rate. Data showed that composed multiple IVs, proteoglycans. The dominant laminin family member in all was α5β2γ1. IV trimer capsule (LC) blood vessel (BV) had chain composition α1(IV)2, α2 (IV), whereas ILM α3(IV), α4(IV), α5(IV) composition. data also ratio IVs varied among different types: other is highest LC, followed BV lowest ILM. have been deposited ProteomeXchange identifier PXD001025.
