Biosynthesis of (10R)-juvenile hormone III from farnesoic acid by Aedes aegypti ovary

摘要: Synthesis of (10R)-juvenile hormone III (JH III) outside the corpora allata (CA) was investigated in female Aedes aegypti. Intact females or ligated abdomens blood-fed and sugar-fed synthesized vivo [12-3H]JH III-like molecules from [12-3H]-methyl farnesoate, indicating that an organ(s) abdomen, other than CA, converted methyl farnesoate into JH III. To find out molecules, ovaries, fat bodies, midguts were incubated vitro with [12-3H]methyl synthesis compared by CA. identify tissue(s) having both farnesoic acid transferase epoxidase, enzymes convert III, bodies removed sugar [12-3H]farnesoic acid. Chemical derivatization methoxyhydrin formation followed esterification (+)-α-methoxy- α-trifluoromethyl phenylacetic (MTPA) chloride reversed phase liquid chromatography identified (10R)-JH (+)-MTPA ester as sole molecule produced tissue culture incubation ovaries. Since only is not racemic oxidation must be enzymatically mediated. Ovaries A. aegypti also [3H,14C]JH L-[methyl-3H]methionine [14C]acetate which characterized HPLC gas chromatography. These results suggest mosquito ovary can synthesize acid, this synthesizes L-methionine acetate. © 1994 Wiley-Liss, Inc.
