摘要: To investigate the role of Epstein-Barr virus (EBV) in epithelial tumors, we compared expression pattern cellular genes EBV-infected gastric carcinoma cell line, NU-GC-3, and its uninfected control. Subtractive suppression hybridization (SSH) was combined with high-density DNA array screening to identify differentially expressed genes. We have discovered that EBV infection upregulated a truncated variant human basic hair keratin 1 (hHb1-ΔN), gene had previously been identified metastatic breast carcinoma. verified differential hHb1-ΔN 3 independent EBV-positive -negative NU-GC-3 clones by Northern blotting. further EBV-dependent upregulation other lines (AGS, TWO3 DLD1) RT-PCR. Inhibition CpG methylation 5-Aza-CdR induced mRNA EBV-negative but did not alter clones. The protein detectable immunofluorescence Western blotting after proteasome inhibitor (MG132) treatment. formed fibrous structures cytoplasm accumulated distinct nuclear bodies euchromatic areas nucleus. suggest unstable may inhibit some functions cytoskeleton and/or interfere transcription regulation.
