Knockdown of DGCR5 enhances the radiosensitivity of human laryngeal carcinoma cells via inducing miR‐195

摘要: Long noncoding RNAs (lncRNAs) exert critical roles in the development of various cancers, including human laryngeal cancer. Radioresistance contributes to predominant causes cancer recurrence after radiotherapy. The aim our study was investigate association dysregulated lncRNA and radiation resistance larynx squamous carcinoma. Here, we investigated biological DiGeorge syndrome region gene 5 (DGCR5) radioresistance Two carcinoma cell lines (Hep-2 Hep-2R), with different radiosensitivities vitro were used present study. We observed that DGCR5 significantly upregulated Hep-2R cells. Inhibition by LV-shDGCR5 transfection restrained proliferation sensitized cells radiation. Reversely, overexpression exhibited an opposite phenomenon vitro. In addition, microRNA (miR)-195 predicted as a direct downstream target DGCR5. Dual-luciferase reporter RNA immunoprecipitation assays verified interaction between them. Meanwhile, miR-195 be reduced mimics repressed Hep-2 growth. Moreover, radiosensitivity greatly enhanced miR-195, which could reversed upregulation Finally, vivo experiments validate knockdown suppressed via targeting miR-195. conclusion, indicated contribute sponging
