摘要: Key components of membrane trafficking and signaling machinery in eukaryotic cells are proteins that bind or synthesize phosphoinositides. PIKfyve, a product an evolutionarily conserved single-copy gene has both these features. It binds to phosphatidylinositol (PtdIns)3P synthesizes PtdIns(3,5)P2 PtdIns5P. Molecular functions PIKfyve elusive but recent advances consistent with key role the course endosomal transport. dysfunction induces endosome enlargement profound cytoplasmic vacuolation, likely as result impaired normal processing exit out endosomes. Multicellular organisms genetically function protein partners regulating homeostasis display severe disorders, including embryonic/perinatal death. This review describes on functionality higher eukaryotes, particular reference biochemical genetic insights partners.
