Phase II study. Concurrent chemotherapy and radiotherapy with nitroglycerin in locally advanced non-small cell lung cancer.

摘要: Abstract Background Nitroglycerin, a nitric oxide donor agent, reduces the expression of hypoxia-inducible factor-1α (HIF-1α) and could be normalizer tumor microenvironment. Both factors are associated with chemo-radio-resistance. The aim this study was to determine safety profile efficacy nitroglycerin administration chemo-radiotherapy in patients locally advanced non-small cell lung cancer (NSCLC). Methods This is phase II trial NSCLC treated cisplatin vinorelbine plus concurrent radiotherapy. A 25-mg NTG patch administered for 5days (1day before 4days after chemotherapy induction consolidation) all day during chemo-radiotherapy. VEGF plasmatic level determined two cycles chemotherapy. Results Thirty-five were enrolled trial. Sixty-three percent achieved an overall response chemotherapy, 75% median progression-free survival 13.5months (95% CI, 8.8–18.2), 26.9months 15.3–38.5). Reduction better OS. toxicity related included headache (20%) hypotension (2.9%). Conclusions addition chemoradiotherapy has acceptable supports possibility add randomized warranted confirm these findings.
