Method for procuring specific populations of viable human prostate cells for research.

摘要: A wider range of research can be conducted on viable tissue samples than fixed or frozen samples. major obstacle to studying prostate is the inability accurately identify cancer direct examination unembedded tissue. We used a dissecting microscope in unfixed stained cut surface with 0.5% aqueous toluidine blue. measured diagnostic accuracy this technique 25 consecutive prostatectomies, determined viability procured samples, and estimated effect final pathologic assessment. Both surfaces 3- 5-mm thick cross-section taken midway between base apex were examined. 4-mm punch biopsy was directed one benign malignant area when clearly present. The allowed clearcut recognition carcinoma 17 cross-sections, confirmed all (100%). In 8 cases, no procurement attempted because evident studied. Twenty cross-sections adequate for procurement; five not suitable presence extensive atrophy. Seventeen 20 diagnosed as (85%); showed pseudohyperplastic adenocarcinoma, focal high-grade prostatic intraepithelial neoplasia, urothelial situ. Prostatic epithelium obtained remains separated from stroma. appears facilitate rather interfere accurate assessment: extraprostatic extension positive margins correctly identified during three cases. procedure takes only about 30 minutes.
