摘要: Cigarette smoke can increase oxidative DNA damage. The main component in cigarette is nicotine. Nicotine metabolized to cotinine, which be regarded as a biomarker for measuring exposure tobacco smoke. A sensitive, simple, and robust method based on on-line solid-phase extraction liquid chromatography with tandem mass spectrometry (on-line SPE LC-MS/MS) has been developed validated the simultaneous determination of 8-OHdG cotinine. matrix effects cotinine were measured at 97.1 91.7 %, values CV 4.4 4.2 %, respectively. limits detection 10.0 5.5 pg mL(-1), quantification 40.0 20.0 pg mL(-1), total run time was 12 min. We quantified urine 80 male subjects. results showed levels smokers significantly higher than that non-smokers. Furthermore, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol its glucuronide conjugate (defined NNAL) are nitrosation metabolites In this study, urinary well correlated NNAL. This also first study focus future risk stress from relationship between levels, NNAL concentrations humans. Graphical Abstract On-line LC-MS/MS human urine.
