摘要: Membrane integrity has been used as a criterion for the definition of cell viability. In present work, staining conditions (time and dye concentration) evaluation membrane in fluorescence microplate reader, using membrane-impermeant nucleic-acid SYTOX Green, were optimized. Incubating Pseudokirchneriella subcapitata algal cells with 0.5 μmol/l Green 40 min allowed clear discrimination between live (intact plasma membrane) dead (with compromised membrane). Algal suspensions, labelled exhibited green proportional to fraction permeabilized membrane. The optimized assess toxicity 1-pentanol on P. short-term exposure (6 h) assay. loss population increased concentration 1-pentanol. 6-h EC10 EC50 values 7,617 mg/l (95 % confidence limits 4,670–9,327) 12,818 mg/l 10,929–15,183), respectively. developed microplate-based assay can be useful high-throughput screening toxics or environmental samples alga subcapitata.
