Morpho-functional characterization of cultured pigment cells from Rana esculenta L. liver.

摘要: A simple method to isolate and culture liver pigment cells fromRana esculenta L. is described which utilizes a pronase digestion of perfused liver, followed by sedimentation on Ficoll gradient. first characterization isolated cultured also reported. They show both positivity for nonspecific esterases, phagocytosis ability, like the phagocytic lineage. Furthermore, after stimulation with phorbol ester, these generate superoxide anions. At phase contrast microscope, present variability in size, morphology, their content dark-brown granules. Inasmuch as cell extract obtained from exhibits specific protein band dopa-oxidase activity, when run nondenaturing polyacrylamide gel electrophoresis, should not be considered melanophages, but that can actively synthesize melanin. The presented here seems useful more directly investigate this extra-cutaneous melanin-containing system clarify its physiologic relevance.
