摘要: Matrine is one of the main bioactive alkaloids Sophora flavescens Aiton, which has been widely used to treat various diseases in China. These include viral hepatitis, liver fibrosis, cardiac arrhythmia, skin diseases, and tumors. However, matrine also toxic compound this herb, available biomarkers are not reliable detecting or quantifying risk. Metabolomics a powerful tool identify early toxicity that specific indicators damage biosystems. This study aimed find potential matrine-induced effects rats HepG2 cells. The toxicological induced by could be derived from elevated taurine trimethylamine N-oxide levels depletion hippurate tricarboxylic acid cycle intermediates, such as 2-oxoglutarate, citrate, succinate urine. Cell metabolomics revealed alanine, choline, glutathione, lactate, phosphocholine, cholesterol showed dose-dependent decreases, whereas taurine, fatty acid, unsaturated increases. Overall, significant perturbation metabolites response high dose was observed both vivo vitro, selected particularly represent an attractive marker for toxicity.
