摘要: We investigated the impact of treatment with parathyroid hormone (PTH) and dexamethasone (DEX) for 2 24h by RNA sequencing miRNAs in primary human bone (HOB) cells. A total 207 million reads were obtained, normalized absolute expression retrieved 373 most abundant miRNAs. In naive control cells, 7 differentially expressed (FDR<0.05) between two time points. Ten exhibited differential (FDR <0.05) across points treatments after adjusting controls selected downstream analyses. Results show significant effects on miRNA when comparing PTH DEX at 2h even more pronounced 24h. Interestingly, several exhibiting differences are predicted to target genes involved metabolism e.g. miR-30c2, miR-203 miR-205 targeting RUNX2, miR-320 β-catenin (CTNNB1) mRNA expression. CTNNB1and RUNX2 levels decreased increased treatment. Our analysis also identified putative novel treated cells showed that affect HOB regulated turn correlated key metabolism.
