摘要: The Freund's adjuvant-injected rat shares a number of features with the arthritis patient, viz presence proliferative synovitis, joint swelling, and cartilage bone erosion. Naproxen, prostaglandin synthetase inhibitor which is an effective antiinflammatory agent in laboratory animals humans, was evaluated as connective tissue destruction this model by use radiologic histopathologic analyses. Sixteen days after rats were injected complete adjuvant, marked swelling noted. On day 17, vehicle or naproxen, 7 mg/kg/day, administered orally. Twenty-eight later vehicle-treated demonstrated following pathologic changes their hindpaws; loss, large amounts pannus within spaces, osteoporosis, erosions, periosteal new formation, heterotopic ossification, bony ankylosis. Rats treated 28 naproxen had significantly milder disease than controls. incidence severe juxtaarticular 10/10 controls versus 2/10 drug-treated group (P less 0.01). A comparable reduction erosion, pannus, formation noted group. These effects may relate to inhibition biosynthesis; prostaglandins have been shown to: 1) stimulate collagenase secretion from macrophages, 2) resorption vivo vitro, 3) diminish proteoglycan synthesis cartilage.
