摘要: HER2 has been found to be amplified in 10-20% of gastric cancers, and is correlated with poor outcome. The aims this study were recognize amplification cancer cell lines via fluorescence situ hybridization evaluate the growth inhibitory effect trastuzumab HER2-amplified lines. To elucidate mechanism inhibition, we performed cycle analysis immunoblotting downstream molecules. We also conducted drug interaction studies other chemotherapeutic agents. was newly identified only SNU-216 cells, moderately inhibited cells positive controls. Trastuzumab-mediated G1 arrest occurred increased expression p27(KIP1) decreased cyclins. Phosphorylation molecules, STAT3, AKT, ERK, by trastuzumab. Treatment plus cisplatin resulted a synergistic effect, whereas treatment 5-FU, or oxaliplatin produced an additive effect. These results suggest that combined agents can active against amplification.
