Original Article Regulation of apoptosis by long non-coding RNA HIF1A-AS1 in VSMCs: implications for TAA pathogenesis

摘要: Objective: Long non-coding RNAs (lncRNAs) play important roles in diverse biological processes, such as transcriptional regulation, cell growth and tumorigenesis. However, little was known about whether lncRNA HIF 1 alpha-antisense RNA (HIF1a-AS1) regulating the proliferation apoptosis of VSMCs vitro expres- sion HIF1a-AS1 serum TAA patients. Methods: The viability detected by CCK8 assay. assessed annexin V-PI double-labeling staining. Expression genes proteins were analyzed real-time PCR western blotting respectively. Cells transfected with siRNAs a gene silencing methods. Results: In patients, expression significantly increased (superior to 6 folds) compared normal control. Moreover, PA induced time- dose-dependent manner, proportion apoptotic cells had gained untreatment group. also upregulation HIF1a-AS1. We found that transfection siRNA decreased caspase3 caspase8 Bcl2, protected PA-induced VSMCs. Conclusions: overexpressed thoracoabdominal aorta aneurysm interaction between plays key role vitro, which may contribute pathogenesis aneurysm.
