Antihepatoma activity of Physalis angulata and P. peruviana extracts and their effects on apoptosis in human Hep G2 cells.

摘要: Physalis angulata and P. peruviana are herbs widely used in folk medicine. In this study, the aqueous ethanol extracts prepared from whole plant of these species were evaluated for their antihepatoma activity. Using XTT assay, three human hepatoma cells, namely Hep G2, 3B PLC/PRF/5 tested. The results showed that extract (EEPP) possessed lowest IC50 value against G2 cells. Interestingly, all no cytotoxic effect on normal mouse liver Treatment with carbonyl cyanide m-chlorophenyl hydrazone, a protonophore, caused reduction membrane potential (Deltapsim) by mitochondrial depolarization. At high concentrations, EEPP was shown to induce cell cycle arrest apoptosis through dysfunction as demonstrated following observations: (i) induced collapse Deltapsim depletion glutathione content dose dependent manner; (ii) pretreatment antioxidant (1.0 microg/ml vitamin E) protected cells EEPP-induced release ROS; (iii) at concentrations 10 50 microg/ml, displayed dose-dependent accumulation Sub-G1 peak (hypoploid) G0/G1-phase arrest. Apoptosis elicited when treated characterized appearance phosphatidylserine outer surface plasma membrane. conclude possesses potent activity its is associated dysfunction.
