摘要: Numerous studies have analyzed the expression and prognostic importance of various proteins in acute myelogenous leukemia (AML). We sought to determine whether sample source methodology used measure protein affect results obtained. To source, we Western blotting compare eight phosphoproteins blast-enriched fraction 118 blood- 108 marrow-derived samples, including 37 paired samples. methodology, five was measured 20 samples by blotting, laser scanning cytometry (LSC), flow cytometry. The mean range were statistically identical for all proteins. Expression measurements blood marrow also had very high statistical correlation. LSC data highest concordance when compared using Kolmogorov-Smirnoff D-stats (range R values, 0.8-1.0). High observed between percentage cells positive dichotomized into or negative expression. However, there less correlation actual percentages compared. majority discordant situations involved that but LSC. signal intensity expression-positive varied limited little heterogeneity either method. In conclusion, provided fractions analyzed, There good LSC, which share similar technology, more these methods blotting.
