摘要: Translocation measurements of intact DNA strands with the ion channel α-hemolysin (α-HL) are limited to single-stranded (ssDNA) experiments as dimensions prevent double-stranded (dsDNA) translocation; however, if a short oligodeoxynucleotide is used interrogate longer ssDNA strand, it possible unzip duplex region when captured in α-HL vestibule, allowing strand translocate through channel. This unzipping process has characteristic duration based on stability duplex. Here, recordings detect presence and relative location oxidized damage site 8-oxo-7,8-dihydroguanine (OG) sequence-specific manner.
