摘要: Publisher Summary This chapter describes procedures to isolate and assay the H + - ATPase from red beet tonoplast. Intact vacuoles isolated storage tissue have been important in establishing major characteristics of tonoplast -ATPase, whereas membrane vesicles homogenates can be prepared large quantities useful characterizing transport catalyzed by this -ATPase. The reviews both methods preparation illustrates salient properties -ATPase are similar those described for other higher plant -ATPases. Red beets obtained commercially or grown field greenhouse. If purchased, care must taken ensure that they were freshly harvested as densities shift upon prolonged storage. All buffers, glassware, rotors cooled 4° homogenate is maintained on ice throughout procedure. peeled quartered before homogenization. method originally devised isolation intact beet, relied use a slicing machine rapidly processed amounts tissue.
