Demonstration of the mineralocorticoid hormone receptor and action in human leukemic cell lines.

摘要: We studied the expression of mineralocorticoid receptor (MCR), and amiloride-sensitive sodium channel (ASSC) regulated by MCR, in human leukemic cell lines. Cell extracts from TF1 (proerythroblastic), HEL (human erythroblastic leukemia) U937 (myeloblastic) line were positive for ASSC, as a 82 kDa band Western blots developed with aid polyclonal antibody raised against peptide QGLGKGDKREEQGL, corresponding to region 44-58 alpha subunit epithelial (ENaC) cloned rat colon, linked KLH. The MCR revealed single about 102 cells. immunofluorescent labelling all lines showed nucleocytoplasmic localization but ASSC was exclusively membrane-bound these results confirmed confocal microscopy. cells evident predicted 843 bp (234 amino acids) electrophoresis PCR product obtained after total RNA had been reverse transcribed then amplified using primers 5'-AGGCTACCACAGTCTCCCTG-3' 5'-GCAGTGTAAAATCTCCAGTC-3' (sense antisense, respectively).
