Topoisomerase II-reactive chemotherapeutic drugs induce apoptosis in thymocytes.

摘要: The effects of topoisomerase II-reactive epipodophyllotoxins etoposide and teniposide as well amsacrine on the viability thymocytes in primary culture has been examined. All three drugs were shown to produce DNA cleavage detectable by resolving isolated pulsed field agarose gel electrophoresis. was found have two components. first due interaction with II, whereas second component endonuclease caused drug-induced entry into programmed cell death or apoptosis. This also detected undergoing apoptosis following exposure glucocorticoid analogue, dexamethasone. effect is dependent upon new protein RNA synthesis, indicating that II a role very stages process. These results are discussed terms use this class chemotherapy.
