摘要: Neuroendocrine tumours (NETs) are uncommon which have a diverse biology. The aims of this thesis were to identify potential new biomarkers and further develop understanding tumour biology in NETs. following assessed i) somatostatin receptor (SSTR) dopamine-2 (D2R) expression NETs, ii)HER their associated prognosis, iii) Angiopoietin NETs prognostic significance, iv) proteomic analysis serum NET cell lines novel markers finally v) the role 68Ga-DOTATATE PET imaging NETs. Immunohistochemical studies performed determine whether SSTR D2R co-expressed was with SSTR-2 -5 93% low grade tumours, lower co-expression higher tumours. HER family receptors involved oncogenesis; expressions these assessed. Immunohistochemical 82 cases. EGFR expressed 86%, HER-2 0%, HER-3 8.5% HER-4 91.5%. not poor prognosis. Angiopoietins (Ang) tumourogenesis. Serum Ang-1 Ang-2 measured patients healthy controls. significantly compared Patients levels >4756pg/ml had shorter time progression. Proteomic using gel electrophoresis LC/MS/MS plasma from established biomarkers. Proteomic line identified 17 proteins all including Mac-2 binding protein. We validated protein it appears be marker for NETs. Finally, we study ascertain identifies more lesions whom 111In-DTPA-Octreotide showed faint/negative lesion uptake. scintigraphy 27 168 Ga-68-DOTATATE PET. is sensitive modality identifying
