A highly efficient extraction protocol for magnetic particles on a digital microfluidic chip

摘要: We present a novel, simple and highly efficient protocol for the extraction of magnetic particles out individual droplets on digital lab-on-chip in presence spatially fixed magnet with permanent field. In this approach, were extracted from droplet by interplay capillary, electrowetting forces, thereby avoiding use mechanical components that would be needed removing when particle resuspension is required. This manipulation allowed execution very fast washing protocols microfluidic (DMF) platform. To demonstrate effectiveness protocol, an IgG immunoassay was implemented DMF Our improved reduced overall assay variability to 3% coefficient variation (CV) while all incubation steps automatically performed on-chip. addition, suspended introduction mixing strategy using as stirrers, resulting improvement 90% detection limit compared passive strategy, solely based diffusion.
