摘要: Abstract The mouse genome contains a retrovirus-like sequence, designated VL30, which is expressed at high levels in transformed cells and can be induced by exogenously supplied epidermal growth factor (EGF). Binding of EGF to the receptor produces changes intracellular calcium phospholipase activity indirectly lead activation protein kinase C. We treated AKR-2B cells, Swiss 3T3 variants NR6 (EGF receptorless) TNR9 (phorbol ester nonresponsive) with various phorbol tumor promoters synthetic diacylglycerol sn-1,2-dioctanoylglycerol. Tumor-promoting esters (e.g. 12-O-tetradecanoyl acetate (TPA] increased level VL30 expression. Stimulation either TPA or produced similar time course expression EGF-receptorless cell line, indicating that neither ligand-receptor binding nor phosphorylation was required for induction Protein synthesis not TPA-mediated increase expression, as pretreatment cycloheximide did block reduce effect. also stimulated treatment sn-1,2-dioctanoylglycerol, an analog probable endogenous activator These results suggest C plays direct role regulating
