Increase in plasma endotoxin concentrations and the expression of Toll-like receptors and suppressor of cytokine signaling-3 in mononuclear cells after a high-fat, high-carbohydrate meal: implications for insulin resistance.

摘要: Objective: To compare effect of a high fat carbohydrate meal (HFHC) with that fiber and fruit on the concentrations endotoxin (LPS), lipopolysaccharide binding protein (LBP), expression toll like receptors (TLR) suppressor cytokine signaling-3 (SOCS-3) in mononuclear cells (MNC). Research Design Methods: Healthy lean subjects were given 910 Calories either HFHC (n=10) or an American Heart Association (AHA) recommended rich following overnight fast. Blood was collected before at 1h, 2h 3h after meal. Cellular indices oxidative inflammatory stress, SOCS-3, TLR2 TLR4 MNC plasma LPS LBP measured. Results: intake induced increase concentration protein, reactive oxygen species (ROS) generation nuclear factor κ B (NFκB) activity ( P <0.05, for all). These increases totally absent AHA fruit. Conclusions: The novel changes described elucidate further mechanisms underlying post prandial inflammation also provide first evidence explaining pathogenesis insulin leptin resistance mediated by SOCS-3 such meals. In contrast does not induce these effects.
