摘要: Background Paracetamol (APAP) hepatotoxicity remains the leading cause of drug-induced liver failure. Fish oil, which contains ω-3 fatty acids, has demonstrated therapeutic efficacy in several models disease. Evidence for its use APAP intoxication, however, is conflicting. The effects fish oil supplementation on APAP-induced failure were investigated. Methods Ten C57BL6/J mice fed a diet based menhaden (MEN) or soybean (SOY) 3 weeks followed by intoxication. In second experiment, prefeeding period was reduced to 5 days. third 10 received study diets weeks, after they chronic, low-dose administration another 4 weeks. Finally, oral parenteral nutrition supplemented with either intravenous (IV) soybean-based oil-based lipid emulsion 19 days, Results extent hepatocellular necrosis (3.8 ± 0.2 vs 2.8 0.2; P = .021) and serum alanine aminotransferase values (2807 785 554 141 IU/L; .048) significantly elevated MEN compared SOY-diet controls. Long-term, did not lead injury irrespective diet. Pretreatment soybean- IV emulsions intoxication no significant differences hepatic between groups. Conclusion Within ranges, harmless dietary fat composition. increase hepatotoxicity, but animals more susceptible, rather than resistant, hepatotoxicity.
