摘要: Dimethyl sulfoxide (DMSO), an amphipathic molecule, is widely used not only as a solvent for water-insoluble substances but also cryopreservant various types of cells. Exposure to DMSO sometimes causes unexpected changes in cell fates. Because mammalian development and cellular differentiation are controlled epigenetically by DNA methylation histone modifications, likely affects the epigenetic system. The effects on transcription three major methyltransferases (Dnmts) five well-studied modification enzymes were examined mouse embryonic stem cells embryoid bodies (EBs) reverse transcription-polymerase chain reaction. Addition (0.02%-1.0%) EBs culture induced increase Dnmt3a mRNA levels with increasing dosage. Increased expression two subtypes protein was confirmed Western blotting. Southern blot analysis revealed that caused hypermethylation kinds repetitive sequences EBs. Furthermore, restriction landmark genomic scanning, which status can be analyzed thousands loci genic regions, affected at multiple loci, inducing hypomethylation well depending loci.
