摘要: We have previously reported that interferon-alpha-2b (IFN-α-2b) can be encapsulated in liposomes without compromising its anti-fibrogenic effects on dermal fibroblasts vitro. This study was conducted to determine whether this preparation applied topically guinea pig wounds affect their healing. The rationale for approach is systemic administration of IFN-α-2b by injection treatment fibrosis uncomfortable, requires a large quantity the cytokine and cannot easily used children. Liposomes are potentially useful vehicles topical delivery drugs. Empty sonicated liposome vesicles were mixed with various concentrations then dried rehydrated. An enzyme-linked immunosorbent assay (ELISA) efficiency encapsulation stability under experimental conditions. A total 36 full thickness skin (6/animal, 3 each side) made an 8 mm disposable punch. Each wound right side received cream (100 mg/wound) containing 3000 units liposome-encapsulated IFN-α-2b, while left empty liposomes. There significant reduction rate contraction treated as early 5 days after wounding. remained up 10 days. Northern analysis, evaluate expression mRNAs type I III collagens response showed marked abundance transcripts pro-α1(I) chain 1 collagen 11 14 Similarly, level mRNA procollagen markedly reduced day 7 depressed 14. These findings consistent results obtained content tissue samples. Cellularity IFN-α-2b-treated wounds, assessed vimentin content, also at Liposome associated completion epithelialization collagen., confirming transepidermal penetration effectiveness. activity liposome-associated vivo supports concept use agent fibroproliferative disorders.
