A Novel Model of Chronic Kidney Disease in Rats: Dietary Adenine in Combination with Unilateral Nephrectomy.

摘要: Background Adenine at 0.75% in the diet (AD) triggers renal impairment rats. This model of kidney disease is largely reversible when AD feeding stopped. Testing novel drugs parallel to administration may result unwanted interference. Objectives We hypothesized that combining with unilateral nephrectomy (UNx) would progressive chronic (CKD) even after cessation AD. Methods In an explorative study, 16 rats UNx (AD-1K rats) and 10 sham-operated (AD-2K received AD-supplemented feed for 3 weeks, followed by 4 weeks standard chow. Ten receiving only chow served as controls. Laboratory parameters blood urine were frequently assessed during feeding. Comprehensive pathological examinations performed all end experiment. Results Rats more affected impaired glomerular filtration rate, anemia, hyperphosphatemia, hypocalcemia. After feeding, recovery was poorest AD-1K rats, paralleled increased proteinuria indicative CKD.
