The neurotoxic effect of 24-hydroxycholesterol on SH-SY5Y human neuroblastoma cells

摘要: Neurodegenerative disorders are characterized by a massive loss of nerve cells. The neuronal cell death is accompanied an increased cholesterol release and conversion into the polar metabolite, 24-hydroxycholesterol (24-OH-Chol), appears to be important mechanism in central nervous system for eliminating from brain. We tested influence 24-OH-Chol on SH-SY5Y human neuroblastoma cells recording morphology, Trypan blue exclusion, LDH-release culture medium, intracellular calcium reactive oxygen species (ROS). exposure 50 microM led 90% viability within 30 h, medium rapidly after 24 h we found elevation calcium. These results show that, physiological concentration range, damages cells, thus speculate that this oxysterol may involved etiology neurodegenerative disease.
