摘要: We demonstrate that single-nucleotide variations in a DNA sequence can be detected using capillary electrophoresis (CE) and molecular beacons (MBs). In this method, the region surrounding site of nucleotide variation was amplified polymerase chain reaction, then hybridize PCR products with each MBs. The sequences are different at 2,044 exon interleukin (IL)-13 which to identified. Through denaturation, product became single strand hybridized completely complementary MB. MB-target duplexes were separated CE solution-based fluorescence techniques. results show reaction fluorescent response elicited from beacon perfectly DNA, but not other MB whose probe mismatched target sequence. method based on MBs is able identify discriminate genotyping SNP between homo- heteroduplexes fragments.
