Functional Analysis and Tissue-Specific Expression of Drosophila Na+,K+-ATPase Subunits

摘要: We have previously purified and characterized a nervous system-specific glycoprotein antigen from adult Drosophila heads, designated Nervana [nerve (NRV)] identified two separate genes coding for three different proteins. All proteins share homology with the β subunits of Na + ,K -ATPase various other species. In this study we isolated new α subunit cDNA clone (PSa; GenBank accession no. AF044974) demonstrate expression functional activity when PSα mRNA is coinjected into Xenopus oocytes along any Nrv mRNAs. Western blotting, RNase protection assays, immunocytochemical staining fly sections indicate that NRV2 expressed primarily in system. Staining most intense brain thoracic ganglia likely associated neuronal elements. NRV1 more broadly muscle excretory tissue also shows diffuse distribution Similar to species, expresses multiple isoforms tissue- cell type-specific pattern. It will now be possible use advantages molecular classical genetics investigate phenotypic consequences altering types.
