摘要: Aseptic loosening of artificial hip joint is a major complication affecting the long-term use joint, and main cause replacement failure. However, mechanism aseptic THR has not yet cleared. The aim this study was to investigate underlying DANCR in osteoblast differentiation (OD). We detected expressions FOXO1 clinical samples mesenchymal stem cells (MSCs) by qRT-PCR western blotting. The effects polymethylmethacrylate (PMMA) on OD MSCs were examined alkaline phosphatase (ALP) activity Alizarin Red S (ARS) staining. markers measured RNA pull-down, immunoprecipitation (RIP) assay ubiquitination assays. Compared with surrounding normal tissues, expression up-regulated down-regulated periprosthetic tissues. PMMA suppressed ALP activity, increased expression, decreased FOXO1, Runx2, Osterix (Ostx) osteocalcin (OCN). ARS staining showed that inhibited MSCs. Knockdown attenuated inhibitory effect OD. could reverse si-DANC. pull-down RIP implicated bound FOXO1. Ubiquitination indicated si-DANCR repress Skp2-mediated LncRNA inhibit regulating expression.
