摘要: Abstract As a tool in genetic engineering, isolated microspore cultures have the remarkable quality to produce doubled haploids for plant breeding and gene mapping. Protocols were developed induction of microspore-derived embryos generation haploid plants from microspores Brussels sprouts. A cytological analysis showed that two modes sprouts embryogenesis exist: direct route via which was major developmental pathway, an indirect calli. Cold pretreatment (4 °C) aminoethoxyvinylglycine addition tested determine whether how they could improve embryogenesis. improved viability microspores, stimulated directly. Similarly, had positive effect on number produced improving multinucleate structures further develop. The combination cold 24 h 1 μM significantly enhanced efficiency, especially with low responsive genotype ‘1076’ it increased by about 82-fold. Subsequently, use solid B5 medium 1% agarose 2% sucrose (w/v) achieved efficient rate regeneration, more than 67% regenerated spontaneous diploids.
