Biosynthesis of Somatostatin-Like Immunoreactivity by Frog Retinas In Vitro

摘要: Somatostatin-like immunoreactivity has been localized to a wide variety of central nervous system neurons, including the retina. We utilized unique advantages retina provides for in vitro studies nerves examine biosynthesis somatostatin. Extracts frog retinas pulse-labeled with [35S]cysteine various time periods revealed uptake radioactivity into material adsorbable by anti-somatostatin antibody linked affinity beads. This increased curvilinear fashion 4 h and was inhibited cycloheximide (0.2 mM) or boiling prior labeling. Pulse-chase experiments that affinity-adsorbable from retinal extracts decreased incubation chase medium; 89% this decrease could be accounted medium. Chromatography on Sephadex G50 (superfine) four elution peaks, whereas only one peak, co-eluted somatostatin-14, identified Chromatographic patterns incubated medium times showed gradual shift earlier-eluting peaks later ones. These indicate somatostatin occurs vitro. The may useful model further study peptidergic neurons.
