Nuclear localization of DP and E2F transcription factors by heterodimeric partners and retinoblastoma protein family members

摘要: E2F is a family of transcription factors implicated in the regulation genes required for progression through G1 and entry into S phase. The transcriptionally active forms are heterodimers composed one polypeptide encoded by gene DP family. transcriptional activity E2F/DP influenced association with members retinoblastoma tumor suppressor protein (pRb, p107, p130). Here intracellular distribution proteins was investigated transiently transfected Chinese hamster human cells. In cells, DP-1 did not accumulate nucleus unless it coexpressed heterodimeric partners E2F-1, E2F-2, or E2F-3. Domain mapping experiments showed that regions E2F-1 stable two were also nuclear localization DP-1. Unlike -2, -3, E2F-4 DP-2, p107 p130, but pRb, stimulated E2F-4, either alone combination DP-2. These results indicate preferentially associate specific partners, suggest ability to localize contributes activity.
