摘要: The human osteosarcoma cell line MG-63 has been used to study the production of bone-specific protein, osteocalcin. In absence any stimuli, cells secreted very low levels secretion osteocalcin started after a lag time 10-12 h upon 1,25-(OH)2D3 treatment. Osteocalcin was measured at doses as 0.03 nM (fourfold increase, p less than 0.05), and this activity increased further with higher reach plateau 50 nM. transiently from in sparse cultures peak values subconfluent (+/- 40%), two- threefold above obtained for confluent cells. Values average 55.9 +/- 2.0 ng/ml protein per 48 h. A similar behavior is observed receptor concentration under experimental conditions. Bmax (40-60% confluent) reached 50% lower However, affinity not affected by density. also possessed an alkaline phosphatase isoenzyme bone-liver-kidney type that stimulated treatment (two- threefold) inhibited parathyroid hormone (40 nM, -25%, 0.025). PTH PGE2 cAMP dose-dependent manner, but were irresponsive salmon calcitonin. Basal PTH-responsive cyclic AMP modulated Dexamethasone pretreatment (100 h) PTH-dependent failed influence response PGE2.(ABSTRACT TRUNCATED AT 250 WORDS)
