The heterogeneous composition of mitochondrial DNA in somatic hybrid calli and the relatively simple composition of such DNA in regenerated leaves

摘要: Mitochondrial (mt) and chloroplast (ct) DNA were characterized in somatic hybrids obtained by fusion of protoplasts from two species, Nicotiana glauca N. langsdorffii. Three a half months after the fusion, total was extracted 50-100 mg callus leaf tissue. Southern hybridization carried out with several probes organelle DNA. The results revealed that widespread rearrangements mtDNA had occurred majority hybrid calli. Even subclones derived same original generated different patterns rearrangements. In contrast to our for calli, we found leaves contained almost simple parental-type mtDNA, namely, langsdorffii this experiment, few detected. Data cob as probe distinctly showed difference between calli leaves. These suggest there may be correlation organization ability regenerate, since nuclear genomes are known stable. Segregation kinds ctDNA observed more frequently than It also apparent no species chloroplasts mitochondria heterogeneous component organelles.
